Conspirators in a Capital Crime: Co-deletion of p18 and p16/p14/p15 in Glioblastoma Multiforme
نویسندگان
چکیده
Glioblastoma multiforme (GBM) is one of the most dreaded cancer diagnoses due to its poor prognosis and the limited treatment options. Homozygous deletion of the p16/ p14/p15 locus is among the most common genetic alterations in GBM. Two recent studies have shown that deletion and mutation of another INK4 family member, p18, also drives the pathogenesis of GBM. This minireview will discuss the known roles for p18 in the initiation and progression of cancer and suggest opportunities for future studies. [Cancer Res 2008;68(21):8657–60]

Identification of p18 as a Tumor Suppressor Gene in Glioblastoma Multiforme
Genomic alterations leading to aberrant activation of cyclin/ cyclin-dependent kinase (cdk) complexes drive the pathogenesis of many common human tumor types. In the case of glioblastoma multiforme (GBM), these alterations are most commonly due to homozygous deletion of p16 and less commonly due to genomic amplifications of individual genes encoding cyclins or cdks. Here, we describe deletion o...
متن کاملAdvance and Briefs Mechanisms of Inactivation of p14, p15, and p16 Genes in Human Esophageal Squamous Cell Carcinoma
The 9p21 gene cluster, harboring growth suppressive genes p14, p15, and p16, is one of the major aberration hotspots in human cancers. It was shown that p14 and p16 play active roles in the p53 and Rb tumor suppressive pathways, respectively, and p15 is a mediator of the extracellular growth inhibition signals. To elucidate specific targets and aberrations affecting this subchromosomal region, ...
متن کاملImaging, Diagnosis, Prognosis Prognostic Significance of p14, p15, and p16 Inactivation in Malignant Peripheral Nerve Sheath Tumors
Purpose: p14, p15, and p16 are tumor suppressor genes that are located closely at 9p21 and are often coinactivated by genetic or epigenetic alterations. Malignant peripheral nerve sheath tumor (MPNST) is a rare sarcoma with poor prognosis. However, the prognostic implications of inactivation of p14, p15, and p16 in MPNSTs have not been adequately investigated. Here we carried out a genetic, epi...
متن کاملCharacterization of a germ-line deletion, including the entire INK4/ARF locus, in a melanoma-neural system tumor family: identification of ANRIL, an antisense noncoding RNA whose expression coclusters with ARF.
We have previously detected a large germ-line deletion, which included the entire p15/CDKN2B-p16/CDKN2A-p14/ARF gene cluster, in the largest melanoma-neural system tumor (NST) syndrome family known to date by means of heterozygosity mapping based on microsatellite markers. Here, we used gene dose mapping with sequence-tagged site real-time PCR to locate the deletion end points, which were then ...
متن کاملDeletion ofpl6 and pl5 Genes in Brain Tumors1
We have used molecular genetic methods to examine the status of cell cycle-inhibitory genes in human brain tumors. We found that p16 and a neighboring gene, pi5, were often homozygously deleted in glioblastoma multiformes but not in medulloblastomas or ependymomas. The deletions occurred in both primary tumors and their derived xenografts, but no Intragenic mutations In either of the two genes ...
